In the next 100 years, there will probably be cities in space.

Question: How do you read “100” years here?
Do you say “a hundred” years or “one hundred” years?
I mean how native speakers say it?

I would say “in the next hundred years” from your written example. “in the next one hundred years” is also possible. I would not say “in the next a hundred years”. That would sound really strange and awkward.
